Abstract

The invention relates to methods for the global and specific isolation and characterization of the in vivo RNA-Binding Proteome (RBPome) present in a cell. The method is performed on cells that have been exposed to UV-light that serves to selectively cross-link cellular RNAs to their associated protein partners under physiological conditions. The method employs RNA splint-mediated ligation to RNA adapters, to selectively isolate cellular RNAs and their associated protein partners, and purification steps that ensure a higher yield and specificity of proteins considered as RBPs.
