PROTEIN CHEMISTRY

Protein purification: Äkta systems using PrimeView/Unicorn 5.31.

Protein characterisation: SDS/Native/2D gel electrophoresis, Western blotting, basal Mass spectrometry (MALDI-TOF).

Protein-conjugation: biotin, FITC, Alexa Fluor, and dendrimers.

 

IMMUNOCHEMICAL ASSAYS

ELISA: highly experienced in any kind of ELISA assays including of development of ad hoc ELISA assays.

Flow cytometry: Using BD CellQuest and BD FACS CAP software. Bead arrays (i.e. CBA).

Luminex: commercial kits, development of ad hoc assays for measuring cytokines, (auto)antibodies and immunogenicity of pharmaceuticals.

 

CELLULAR

Cell isolation: density-gradient centrifugation; magnetic separation (Dynal bead, MACS); from blood and tissue.

Mammalian cell cultivation: PBMC, bCSC, IPEC-J2, CHO, A547, U87.

Cell stimulation: mononuclear blood cells (PBMCs).

Toxicology: D2C7-IT in cancer cell lines; natural antibodies in neonatal porcine intestinal epithelial cell lines. MTS assay for cell viability. Complement-mediated cytotoxicity in PBMCs.

Confocal microscopy: Zeiss LSM 780 for evaluating D2C7-IT internalisation.

 

MOLECULAR BIOLOGY

PCR: basal and qPCR.

Cloning: sub cloning in E. coli; yeast, CHO and A549 cells.

Expression of recombinant proteins: in E. coli, P. pastoris and CHO cells.

Fermentation: E. coli, using BioFlo 115.

Gene reporter assay: using IFN-beta sensitive A549 cell line for measuring neutralising IFN-beta antibodies.

 

ANIMAL STUDIES

Weaner piglets: testing dietary prebiotic feed supplement, and autopsy.
